Dawkins, arguably the most famous atheist of his generation, shows in his book why people started believing in God and why anything supernatural almost definitely doesn't exist, through the power of Darwinian natural selection. He has written, perhaps, the most powerful and convincing set of arguments against the existence of the supernatural God.
The initial chapters attempt to distinguish between science and religion (He says that science deals with questions of “how,” whereas religion deals with questions of “why”). They also go on to briefly put forward the points theists use to support their side of the argument.
The next few chapters work as an attempt to explain the roots of religion and morality. They also explain why we should be hostile towards religion (Childhood indoctrination, abortion-clinic bombers, punishment of homosexuals, abuse of children by priests).
The final chapters try to discuss the idea of religion filling the role of a consoler, an imaginary friend, or a big brother always looking over your shoulder in times of need. Dawkins asks us to shrug off the evolutionary chains and embrace a scientific outlook that is “liberated by calculation and reason.”
