- Jenn Leitner: Main character, vampire slayer trained at the University Salamanca in Spain, one of two members of the team that are human. Originally from Berkley, California, and is in love with Antonio.
- Antonio de la Cruz: Vampire who becomes a hunter and hides his identity from everyone except his team. He originally wanted to be a Catholic priest, but was turned into a vampire during WWII by Sergio. He is in love Jenn Leitner
- Aurora Abregon: Vampire royalty, her sire is the same an Antonio's, is part of a master plan that involves her sire and Antonio, evil to the core.
- Skye York: White witch from an old, revered family of witches. Salamancan hunter team member, hiding a dark secret from her team members
- Father Juan: Catholic priest at the University Salamanca, spiritual advisor and leader of the Salmancan Vampire Slayers.
- Heather Leitner: Jenn's sister who desprately wants to be a hunter like her sister to leave her vampire filled town.
- Eriko: Japanese student who becomes "the Hunter" after finishing her training at Salamanca, the leader of the Salamancan Vampire Hunting team, gains supernatural powers after drinking the elixir given to "the Hunter."
- Holgar: Scandanavian member of the Salamancan Vampire Hunting team, is a werewolf. Is the more relaxed member of the group who is always up for laughs
- Jaime: A hotheaded Irish streetfighter who is the second human member of the Salamancan Vampire Hunting team. His family members were killed by werewolves and vampires, which causes friction on the team due to his dislike and mistrust of Holgar and Antonio.
- Solomon: Leader of the vampires who announces to the world from the UN that vampires are real, harmless and want to live in peace and harmony with humans.
- Papa "Che" and Esther Leitner: Jenn's beloved grandparents who inspire her rebel spirit
- Lorien: Vampire lord of San Francisco, partner in evil with Aurora.
- Dr. Michael Sherman: Scientist who invented a virus that will supposedly kill vampires with a strong form of leukemia.
- The Resistance fighters: A rag tag group of New Orleans residents fighting the vampires who help the Salamancan Vampire Hunting team when they arrive in New Orleans.
- Jamie: The other human in the Salamancan Vampire Hunting team whose parents were killed by werewolves. He grew up doing harsh training in Belfast and is skilled streetwise.
- Marc: The leader of the resistance in New Orleans who is very suspicious of anything supernatural
- Alice: Marc's mother who can also do voodoo
- Estefan: Skye's creepy stalking ex-boyfriend that no one knows about
