This is a sequel to Assassin's Creed: Renaissance. Ezio, the master assassin, seeks to avenge the death of his uncle. Unbeknownst to him this will pit him against the Knights Templar.
Ezio Auditore: The main protagonist. An assassin looking for revenge on the ones that got his father and two brothers hung.
La Volpe: The leader of the thieves guild, operating in the city of Venice, though with influence far beyond this. His name is Italian for The Fox, which reflects his nature of stealth when committing crime. La Volpe is shown to be among the most subtle characters, being able to surprise even the assassins with his natural abilities.
Mario Auditore: Mario Auditore is Ezio's uncle, as well as being both his mentor and ally. Mario holds control of the town of Monteriggioni, where his manor home is located.
Cesare Borgia: Cesare Borgia is the main antagonist in the story, and is a real-world figure from the era. Cesare acted as a military general in Italy, allying himself with the French at a point.
Rodrigo Borgia: The real-world Rodrigo Borgia is also known as Pope Alexander VI, and was a Spanish-born religious leader. In the Assassin's Creed franchise he is portrayed as a high-ranking Templar agent, who is searching for an ancient artifact known as the 'Apple of Eden'.
Lucrezia Borgia: Lucrezia Borgia was the real-world sister of Cesare, and daughter of Rodrigo. In the Assassin's Creed franchise she is shown to be cruel and ruthless, though her cruelty does not compare to that of her brother. She is a relatively minor antagonist.
Brotherhood takes place in Italy at the height of the Renaissance.
Rome: As the capital city of Italy, Rome appears during the story as one of the most major cities due to its occupation by the Borgia family, who are shown as the primary antagonists.
Vatican: The Vatican is of extreme importance, since it is here that Pope Alexander VI resides.
Assassin's Guild: Guild that promotes peace and free will in the people. They kill one person rather than slaughter hundreds.
Thieves' Guild: Led by La Volpe from a base centered outside the city, they oppose authority due to their criminal activities. Their flow of information comes from a large number of informants and their spies who observe the Borgia family, as well as their guards.
La Fior la Rosa: Courteans is what is here they work for the Assassin's Guild under the guidance of Ezio's sister Claudia and get information straight from the Cardinals of Roma its self
The Cardinals: Organization that runs the city of Roma under the guidance of the Pope they are loyal followers and will do whatever he says and are like Congress to the U.S
Mercenaries Guild: Led by Bartolomeo, the guild of mercenaries contains well-trained soldiers from across the world, who will work for money. They are, unsurprisingly, opposed to the Borgia family and their advances.
The Eagle: The Eagle is seen as the symbol of the Assassin's Creed, since it reflects their usage of high vantage points and landmarks to strike from, as well as the costume design.
