Diana Bishop is the last of a family of distinguished and well-respected witches. However, she seem reluctant or unable to use her magic which will burst unexpectedly and dangerously from her when she becomes emotional. When seven years old, Diana moved into the home of her Aunt Sarah and her partner, Emily, when her parents were discovered killed under mysterious circumstances while on a research trip to Africa.

While doing historical research at Oxford, the now Dr. Diana Bishop discovers an ancient and enchanted manuscript on alchemy called Ashmole 782, which has been lost for several years. Since Diana discovers it and begins to access its secrets, she finds herself attracting an increasing number of paranormal creatures, including a 1500 year old vampire, Matthew Clairmont. As Diana and Matthew becomes increasingly attracted to each other, the two and their respective families and friends draw the attention of the Congregation, a paranormal governing body comprised of three representatives of witches, vampires and daemons, which prohibit inter-species fraternization. The Congregation is interested in what Diana has learned from the Ashmole 782 and to prevent her from sharing the manuscript or what she has learned with Matthew.

Diana, not fully recognizing the importance of the Ashmole 782, returns it to the book stacks finding herself unable to retrieve it again. Although she noticed that three front pages were torn from the book, she thinks nothing of it until she receives an envelope from her long deceased mother containing one of the three manuscript pages and a letter explaining her inability to exercise her magic.

With Matthew help, Diana becomes more competent in the using and controlling her magic. However, her life is becoming increasingly threatened causing Matthew and Diana to seek refuge in 16th century England.
