- Tsukune Aono: The male protagonist of the series, Tsukune Aono is mistakenly enrolled in an academy for monsters. Though ordinary and often cowardly in the beginning, Tsukune's willingness to protect his friends without thinking twice about it earns him many new companions and the affection of the girls surrounding him.
- Moka Akashiya: Is considered the most beautiful girl at Yōkai Academy and is the first person Tsukune Aono meets when he sets foot in the yōkai realm. Exceptionally sweet and kind, Moka is a vampire, and takes a liking to Tsukune, after ingesting his blood, and becomes addicted in doing so.
- Kurumu Kurono: A Succubus who is considered to be the second most beautiful girl at Yōkai Academy, she is keen on making Tsukune Aono her 'destined one'
- Yukari Sendou: Is one of the smartest students at Yōkai Academy, and the youngest at age 11. She is a witch, and always wears a typical witch costume (consisting of a hat and cape).
- Ruby Toujo: A witch from the human world. Despite not being an official student, she works with the school board.
- Mizore Shirayuki: Frequently seen with a lollipop in her mouth, she has the ability to control the element of ice at will, and can form clones composed of said element. She has developed into a serene, reserved and cute girl with limited emotions.
- Kokoa Shuzen: The younger half sister of Moka. Bearing a similar appearance to Moka, except for her Flameing orange red hair (though it is stated she dyes her hair this colour) and green eyes, she arrives as a first-year student to Yōkai Academy.
